Peroxisome Proliferator-Activated Receptors (PPARs) are a group of nuclear receptor proteins that function as transcription factors to regulate metabolic health. There are three types of PPARs: alpha, gamma and delta. When bound to by fatty acids (such as prostaglandins) influence processes such as fat burning, lipid storage and glucose metabolism. [1]

  • PPAR-alpha serves to metabolise fatty acids, increasing the beneficial HDL cholesterol whilst decreasing triglycerides. [2]
  • PPAR-gamma is the target of medications that boost insulin sensitivity in type 2 diabetes by promoting the storage of fatty acids into adipocytes, and thus correcting dyslipidaemia. [3] These medications, called thiazolidinediones, shift the body in the direction of using glucose as the main fuel substrate rather than free fatty acids. [4]

Among the long list of side effects of Accutane, at the top of the list of immediate concern is an elevation of LDL cholesterol and triglycerides. This puts even young patients at risk of developing cardiovascular disease and atherosclerosis, and so practitioners often require frequent blood tests. [5] [6]
